I think it would be very revealing to see where you are BEFORE the shot.

While the actual serum levels from the injection, typically fall off at 72 hrs, there might still be some floating around, giving a false positive.

This subject is pretty complex. I think you need to carefully read the links we have gathered over time, so that you can understand this situation better. Some homework is inevitable,
and don't assume all doctors are up to date on this subject. Many don't even know what methylcobalamin IS, for example.
On the link I gave you there is a link to American Association of Family Physicians, which is an teaching article on this subject for doctors to read. In it are recommendations to use ORAL B12 supplements. They do not go so far as to explain using on an empty stomach--which is the best way to do orally, but the article is very good otherwise. It is from 2003, and not that "new" anymore.
Table 4 gives suggested dosing.
