Here is a link to a good overview of this product:

Whats the TRUTH about Nicotinamide Riboside (NIAGEN) | Alivebynature - Evidence Based Reviews

Only 12 people were in the study discussed. And of them the only endpoint was serum blood levels, and not metabolism changes since the study participants were "normals" and not sick.

Here is the endpoint of the article:
Quote:

However until more research is published, we don’t know if the benefits seen in studies with mice are confirmed in human testing.
Conclusion: YES, NIAGEN works at NAD+ levels. We just don’t know how much the increased NAD+ levels benefit in restoring youthful function to aging cell.
I think $60 for 60 capsules is a bit much.

I learned at a medical seminar a short while ago that rodent studies only pertain to humans about 60% of the time.

So readers here can make their own decisions. If anyone decides to try this, please keep NT in mind and post your results/progress.

A couple members on another health group I'm a member was just discussing this but I ignored it .. just don't know. I was taking Niacinamide for arthritis but stopped that when I went to the hospital/rehabs.

This is not what I tried. I get no points for reading comprehension.

Nicotinamide and niacinamide are the same thing.
The riboside makes it a different compound from niacinamide
hence a more specific product to make NADH which is the target
desired medically.

Adding to what mrsD cited:

" YES, NIAGEN works at NAD+ levels. We just don’t know how much the increased NAD+ levels benefit in restoring youthful function to aging cell."

This is a reference to what is called a "surrogate outcome" study. It is not a surprise that NIAGEN increases blood NAD+ because it is a biochemical precursor of NAD+. Surrogate outcome studies are often very unreliable as predictors of therapeutic benefits.

Drug companies are fond of surrogate outcome studies because they are quick and cheap to do.

Using intentionally silly words to make my point: A drug company might report "Our WonderDrug77 blocks the action of the Zogbeta4 receptor. Over-active Zogbeta4 receptor is a risk factor for Fred's Encephalopathy.". The inference that is intended to be drawn from this is that the drug is therapeutically beneficial. This does not follow.

A real equivalent of this is that many surrogate outcome studies have shown that statins decrease blood cholesterol levels. This is no surprise because statins inhibit HMGCoA reductase, which catalyses a step in cholesterol biosynthesis. It does not follow from this that statins are, with acceptably low adverse side-effects, useful in treating patients who are at increased risk of cardio-vascular disease because of elevated cholesterol levels.

The contrast with a surrogate outcome study is what is called a "real-life outcome" study. Often drug companies do not like them because they are expensive and take a long time.

A real-life outcome study poses the question "Does drug X, compared to placebo, lead to improved health outcomes with acceptably low adverse side-effects?".

Bottom line; surrogate outcome studies mean very little. Only the results of a well-designed real-life outcome study will help people to make an informed choice about whether a drug may help them.

Gotcha. I purchased Now niacinamide on Amazon for something like $10.

I have used NADH in the past, did help enhance my 'mental acuity", which was subpar after six months of Interferon and Ribavirin treatment.

I'll probably re-order NADH, just trying to use up the supplements I've bought, before getting more. :winky:

Here is my recent experience with Nicotinamide Riboside

I am a 66 year old male with type 2 diabetes, heart disease and high blood pressure. I herniated a disc in the L4/L5 region in 2011, and have reinjured my lower back 6-7 times since, the latest time being early March of 2018. I began taking 125 mg of NR (tru niagen version) on April 1, 2018. April 14, 2018 I began taking 250 mg, and 5/18/18 I increased my dosage to 375 mg. On 6/10/2018 I moved from 375 mg in the morning, pre-breakfast, to 250 mg pre-breakfast and 125 mg post-dinner. Here are the positive side-effects I have noticed:

1) On April 30, 2018, I noticed that 90 percent of my chronic pain/stiffness/discomfort during the day was gone, and about 50 percent of my night time pain/stiffness/discomfort was gone.

2) As I reinitiated an extensive exercise routine, I went from 100 minutes of aerobic exercise per week in mid-April to 400 minutes per week by the end of May. Recovery pain/discomfort was minimal and surprising given the ramp-up.

3) I have noted an increase in flexibility since April, particularly in hips, shoulders, and neck

4) My sleep pattern has stayed the same, but my sleep feels deeper and more restful. When I take a nap, they last 20-40 minutes with the same feeling of refreshment I used to get from a 75-90 minute nap.

5) Spring allergy symptoms are virtually non-existent compared to previous years.

On June 10, noting that I was still feeling some pain/discomfort/stiffness at night in bed, I switched 125 mg NR to post-dinner along with 250 mg of sustained release niacin. This alleviated the majority of the night-time pain/discomfort/stiffness I had been feeling.

I have tried many supplements over the past 40 years and none have had the huge positive impact that NR has had. Highly recommended for anyone over 50 with chronic pain and the desire to lead a more active lifestyle.
