Default Adamas Initiates Phase 3 Trial of Parkinson's Drug

Adamas Pharmaceuticals Inc. announced that it has initiated a pivotal Phase 3 clinical study of ADS-5102 (amantadine HCl), a high-dose, controlled-release version of amantadine for the treatment of patients with Parkinson's disease who have developed levodopa-induced dyskinesia (LID).

Default Question?

Amantadine helped control my dyskinesias somewhat, and mine can be so bad (and painful) that I would welcome anything! However, after my second week on Amantadine, my legs and feet swelled painfully and I had to stop the Amantadine.

Does anyone know if the "new and improved" Amantadine has similar side effects?
Or does anyone know of something else that might be causing the swelling of my lower extremities? I take Sinemet, Comtan, Requip as my regular Parkinson's regime.

Peggy, I have a copy in my office of a presentation Adamas gave on the results for the Phase 2/3 EASED study recently completed. I focused on the efficacy results when I reviewed it and don't recall specific mention of that side effect. I'll take another look at it tomorrow. I've been using amantadine for two years with out any side effects at all. It definitely helps reduce my hand tremor.

Default Peripheral edema

Three volunteers 0f the total group (N = 83) suffered from peripheral edema as an adverse side effect. Unfortunately, I don't know how that compares percentagewise with the original drug formulation. However, although only a small percentage (3.6%), it appears that some people will have lower limb swelling as a side effect.
